Proteinase K Market Overview
Global Proteinase K Market market to grow at a high CAGR Of 7.6% during the forecast period 2023- 2030. The market is expected to grow due to increasing demand from Bio-Pharmaceutical Companies, Pharmaceutical Companies, Academic and Research Centers. The competitive rivalry intensifies with New England Biolabs, Thermo Fisher Scientific Inc.., Merck KGaA, and others operating in the market.
Proteinase K is a typical member of the subtilisin family of proteinases. Proteinase K is able to digest hair (keratin), hence, the name "Proteinase K". The predominant site of cleavage is the peptide bond adjacent to the carboxyl group of aliphatic and aromatic amino acids with blocked alpha amino groups. It is commonly used for its broad specificity. This enzyme belongs to Peptidase family S8 (subtilisin). The molecular weight of Proteinase K is 28,900 daltons (28.9 kDa). Proteinase K is used for the destruction of proteins in cell lysates (tissue, cell culture cells) and for the release of nucleic acids since it very effectively inactivates DNases and RNases.

Proteinase K Industry Dynamics
The Proteinase K market is to witness positive growth owing to higher research, development activities and investments by institutions, pharmaceutical and biopharmaceutical companies in innovation. The growing use of Proteinase K in drug discovery and disease diagnosis will affect the market positively. However, purity issues like the presence of other proteins and nucleases and the low thermal stability of Proteinase K can hamper the market.
Increasing research and development activities will drive the market growth
The increasing adoption of proteinase K in the R&D of rare diseases is one of the major factors that is going to offer lucrative growth opportunities. For instance, in April 2019, researchers from Scripps Research reported that labeling light chains with fluorophores and coupling shape changes to cleavage by proteinase K in order to identify a group of small molecules that prevent structural changes to proteins that that cause AL amyloidosis, a rare and progressive disease.
In drug discovery and disease diagnosis
Fine-needle aspiration biopsy or FNAB is an important tool used in medical research. It is widely adopted, thanks to its simplicity, cost-effectiveness, and safety for diagnosing thyroid disease. Conventionally, the disease is diagnosed with the cytopathological examination. However, newer methods like liquid-based cytology are proving to be more advantageous with more accuracy in results. Proteinase K cells used for RNA extraction in this process are emerging as a key problem-solver for cancer-derived K1 cells and FNAB specimens. The proteinase K is likely to solve challenges relating to RNA isolation and cell recovery in liquid-based cytology. This is a major development in the Proteinase K market and can open tremendous new opportunities for existing players.
The proteinase K products are used in tissue engineering and many other types of research.
Side Effects Associated with the impurity and low thermal stability
The new sophisticated DNA purification technologies can hamper the market. The presence of impurities in proteinase K can affect its use. Low thermal stability of proteinase K limits the hydrolysis and aminolysis reactions in industrial processes, which is expected to hamper the growth of the market over the forecast period. The thermal stability of proteinase K limits its practical potential as an efficient catalyst.
COVID-19 Impact on Proteinase K Market
Direct Clinical Evidence Recommending the Use of Proteinase K or Dithiothreitol to Pretreat Sputum for Detection of SARS-CoV-2. One of the primary tools for diagnosing COVID-19 is the nucleic acid-based real-time reverse transcriptase-polymerase chain reaction (RT-PCR) test performed on respiratory specimens. Proteinase K has great potential in detecting COVID-19. Which is also anticipated to provide growth avenues to the market.
G-Biosciences SalivaOptimize™ Proteinase K has been optimized to provide maximum proteolytic activity within a short period for rapid and maximum release of Covid-19 viral nucleic acid for RT-qPCR detection technique.
Proteinase K Market Segmentation Analysis
By form, the proteinase K market is divided into Lyophilized Powder Form and Liquid Form.
The lyophilized powder form is expected to see growth in the forecast period
Benefits of lyophilized reagents such as proteinase K include high stability, long shelf life and inexpensive shipping. Lyophilization also does not reduce the activity of proteinase K. Lyophilized proteinase K is stable enough to ship at ambient temperatures.
Based on Application, the Proteinase K market is segmented into Purification of DNA & RNA, In situ Hybridization, Mitochondria Isolation and Enzyme Removal
Purification of DNA&RNA segment is seen to witness an increase in the market
Use of Proteinase K in DNA extraction from food samples to know the presence of the living organisms. One such application involves homogenization of frozen northern shrimps in 200 µl of water and blending the homogenates with Lysis Solution CBV and Proteinase K. Thus increasing usage of proteinase K is expected to drive the market growth over the forecast period. The growth of the segment is attributed to the rising application of purified DNA and RNA in various molecular biology processes. These processes often include the development of novel gene therapies which are increasingly being researched across various research institutes.
By End-user, the Proteinase K market is segmented into Bio-Pharmaceutical companies, Pharmaceutical Companies, Academic and Research centers and Contract Research Organization.
Bio-pharmaceutical held the highest position in the proteinase K market
An increasing number of R&D studies for various therapies and innovations with the use of proteinase K will impact the growth of this segment. Apart from these factors, rising demand for recombinant genes for the production of monoclonal antibodies and other biological products from pharmaceutical and biopharmaceutical companies might impact the proteinase K market positively over a period of forecast.
Proteinase K Market Geographical Share
North America will see growth in the market over the forecast period
An increasing number of diseases and polymerase chain reaction applications found will boost the market in this region. Along with increased government funding and more R&D in the life science sector this region will see a rise in the market in the forecast period.
Moreover, with the emergence of COVID-19, the use of proteinase K increased and new products were developed.
According to the Pharmaceutical Research and Manufacturers Association (PhRMA), 2018, US firms conduct over half the world's R&D in pharmaceuticals. Approximately 79.6 billion was invested in biopharmaceutical research and development in the United States in 2018.
